药物亲和反应靶向稳定性技术(Drug Affinity Responsive Target Stability,DARTS)凭借其广泛的适用性和靶点鉴定的稳定性,自2009年首次报道以来,被国内外众多学者和研究团队应用于小分子药物和天然产物的靶点筛选。DARTS技术基于蛋白酶解稳定性的原理。即小分子与蛋白结合后,可能使蛋白的结构发生解旋或聚集,从而使蛋白对广谱的蛋白酶变得更加耐受或敏感。通过SDS-PAGE分离后进行质谱鉴定,即可得到小分子的潜在靶点蛋白。

案例1:人参皂苷Rb1靶向QKI调控外泌体环状RNA,防止结直肠癌肝转移
上海中医药大学曙光医院团队在《Molecular Cancer》上发表了题为“Targeting circ-0034880-enriched tumor extracellular vesicles to impede SPP1highCD206+ pro-tumor macrophages mediated pre-metastatic niche formation in colorectal cancer liver metastasis”的文章。研究发现中药来源的人参皂苷Rb1可显著抑制具有促进癌症转移作用的外泌体circRNA(circ-0034880)表达。通过DARTS技术,研究人员发现了人参皂苷Rb1通过结合并抑制环状RNA生成中的一个关键蛋白QKI发挥作用,并利用SPR等技术实验证明了二者间的直接相互作用。(达吉特为该研究提供了DARTS和SPR技术服务)

案例2:内源性代谢物肌酸靶向朊病毒蛋白PrP影响子宫内膜异位症进展
复旦大学附属妇产科医院研究团队在《Advanced Science》发表题为“Creatine Promotes Endometriosis by Inducing Ferroptosis Resistance via Suppression of PrP”的研究论文。本研究发现子宫内膜异位症患者腹腔液及病灶中肌酸水平显著升高,并提示其主要来源于内源性合成。进一步采用DARTS+MS技术筛选肌酸的作用靶点,发现朊病毒蛋白PrP(PRNP)可与肌酸发生直接相互作用。机制研究表明,肌酸结合PrP并抑制其活性,进而减少细胞铁摄取,降低游离铁及储存铁水平,减弱氧化应激和脂质过氧化,使细胞对铁死亡产生耐受,从而促进子宫内膜异位症进展。该研究通过DARTS技术建立了肌酸与PrP介导的铁稳态调控之间的联系,为理解肌酸参与子宫内膜异位症发生发展的分子机制提供了新的视角。(达吉特为本研究提供了DARTS筛靶技术服务)

案例3:卡格列净通过肠道菌群代谢物靶向乙二醛酶GLO1改善肾损伤
郑州大学附属第一医院团队在《Advanced Science》发表题为“Canagliflozin Alleviates Diabetic Glomerular Endothelial Injury via Melibiose in a Microbiota-Dependent Manner”的研究论文。本研究发现卡格列净通过重塑肠道菌群、上调菌群关键代谢物蜜二糖,而发挥肾保护作用。通过DARTS+MS技术发现蜜二糖直接靶向GLO1,抑制AGE-RAGE通路,从而保护肾小球内皮屏障。为卡格列净的肾保护作用提供了全新的“肠-肾轴”分子机制解释,同时为DKD的治疗提供了全新的先导化合物和直接靶点。(达吉特为本研究提供了DARTS筛靶技术服务)
