药物亲和反应靶向稳定性技术(Drug Affinity Responsive Target Stability,DARTS)是由美国University of California科研团队于2009年提出的。该方法操作简便,可适用于大多数小分子化合物。DARTS技术是基于蛋白的酶解稳定原理,即小分子与蛋白结合后,可能使蛋白的结构发生解旋或聚集,从而使蛋白对广谱的蛋白酶变得更加耐受或敏感。通过SDS-PAGE分离,进行实验组和对照组的条带比较,最后进行质谱鉴定,即可得到小分子的潜在靶点蛋白。

研究案例1:DARTS助力揭示天然产物抗肿瘤的新机制
上海中医药大学张卫东教授团队采用cMAP预测联合DARTS技术,发现水仙环素(Narciclasine)的靶点蛋白STAT3,通过MST与分子对接实验进一步验证了水仙环素与STAT3的直接作用关系。最后通过功能性实验证实了水仙环素能够与STAT3结合发挥抗肿瘤作用。

研究案例2:外泌体+DARTS发现人参皂苷Rb1对结直肠癌的治疗作用
上海中医药大学曙光医院联合了药物靶点和外泌体机制两大研究热点,发现携带circ-0034880的外泌体可以促进结直肠癌的肝转移,随后筛选到了能够显著抑制该环状RNA表达的中药活性成分人参皂苷Rb1。进一步采用DARTS技术鉴定到了人参皂苷Rb1的作用靶点是与环状RNA生物发生相关的蛋白QKI。通过CETSA和SPR方法验证了二者间的相互作用。最后体内功能实验证明了Rb1预处理的肿瘤细胞来源的外泌体失去了促进癌症肝转移的作用。
